Wondering what is the way to get best deal on hotel room block. Looking to reserve 200 rooms at a 3-4 star (Hilton/Hyatt) hotel in Seattle area for an event.

Do hotels give best rates when you work through agents?

For that quantity of rooms I imagine you would be negotiating with the sales manager for the hotel who will put together a contract.

There are also companies that deal with large meetings and such that can go out and negotiate on your behalf. Might start with Visitor bureau.
